I have a 2003 Jeep Wrangler. It has no rear differential vent. I assume it had one and for some reason the previous oner welded over the fitting. Can I simply weld a fitting on the differential cover near the top and attach a vent hose there?

I did't know that, thanks. Brakes are good, but not sure how that works. I don't see anywhere the break T goes into the rear end. Attached is what I have. I'm not the first oner and this may not be the original rearend.

The metal lines intersect at he vent IIRC with a block. That block is attached to the axle tube several inches away from the pumpkin with a hollow bolt and the vent line attached to it and then up to the frame back buy the tank..IIRC. Only comment I have is the vent attached to the cover may be more likely to poo some gear lube out of it due to the lube getting blasted about in the diff. The vent hose should be up as high as you can get it to help. It dont need much of a hole, just enough to allow air to go in and out as the lube and diff heat up and cool down.
Are you 100% certain that there isn't a vent hole up there and maybe just clogged with dirt? It doesn't make sense that somebody would just weld it shut for no reason.
And I see the shadow of the bolt hole through that T. It's essentially a hollow bolt that runs through the T.

The axle vent bolt is still available from mopar or aftermarket as well. The hole in the axle tube is threaded and like everyone said, it holds the brake line T to the axle tube.

OR.. .if you don't mind chinesium... you can get one of these... but I'd really look for the original port first.
The gear oil residue that attracted all that dirt says that the hole is probably still there and just missing the vent bolt.
