Not sure what the PO had in before. I changed to synchromesh @3000 miles ago. All gears and shifting is fine except from 1st to 2nd. If i pull straight down in from 1st to 2nd, 70% of the time, there is a slight grind. If i pull from 1st to neutral, and smack the shifter to the left, then down into 2nd, it wont grind but sometimes there is a light clunk. NV3350. Thoughts?
Would Redline be better for transmission?

I have always seen an improvement using Redline MTL over other oils in the NV3550. How much improvement really depends on the condition of the transmission's internals.
You don't say which brand of synchromesh you're using but GM's and Pennzoil's Synchromesh (both are the same product) reviews are not all positive. I've read of too many complaints about the shifting quality with those two brands of Synchromesh. Plus Synchromesh from GM and Pennzoil is a conventional gear lube which doesn't do much to help the shifting in cold weather.
So while Redine's MTL is not a miracle worker if there's a mechanical problem that needs to be repaired, I'd say you couldn't do better than MTL which is a full synthetic.

In that case I'd say Redline's MTL would definitely be a much better choice for your NV3550.
I have the same issue with my NSG370 6 speed. The first to second shift would give a slight grind. If I gently let it fall in to gear it usually doesn't do it. When I am not thinking about it though and I just pull back to 2nd I feel it. I just changed the fluid over to Redline MTL 2 days ago and didn't notice any change. I think it's a common problem though, I will just have to be more mindful about the shift.
When I was first learning to drive it, I learned that I could either avoid it with the “pause” at the synchro, or by shifting at 3000 RPM. By doing either of those plus the redline, I very rarely grind it anymore.
Go to Redline. You'll notice a difference between Synchromesh and won't regret it at all. AX-15s definitely love it.
And don't waste your time or money on Royal Purple, get the RedlineMTL

Put synchromesh in mine right after I bought it, had same problems you are having and progressively got worse, almost felt like the gears were sticking during shifts. Swapped out to mtl and all is well.
I wouldn't say that, from my experience they perform the same in my tranny in terms of shifting.
Rp or rl, I'd use which ever is cheaper.
After a shop filled my NV3550 with ATF, I drained it and tried the Mopar fluid, Pennzoil, and Redline MTL. I don't remember if the Mopar or Pennzoil was first, but it currently has MTL in it. I haven't noticed much difference in any of them. I get an occasional grind going from 1st to 2nd and sometimes reverse is difficult to select until I let the Jeep roll a bit or pop it into 5th first. I've found if I take an extra fraction of a second shifting, the 1-2 is smooth enough.
