Misfire on new long block out of ideas

I installed a new stroker long block. Started break in just fine and developed a misfire on cylinder 2. I’ve replaced coil, cam position sensor, injectors, plugs, o2 sensor, compression is high. When I replaced the cam positioning sensor I was top dead center on all my marks on compression stroke After driving 5 miles I got a misfire but it jumped to number 5. Can I have a single cylinder misfire if the cam sensor is off? Would bad CAT cause a misfire to move? I’m at a loss.
 
I'd be checking your opda again, only because I had something similar happen. When I checked it after replacing it, I didn't do the clamp tight enough and had bumped at some stage. I then just got the motor to TDC, lined up the opda pin and re tightened the clamp and it was all good 👍.

I redid the opda. I went back to the original one where I had the misfire on cylinder 1. It’s on 5 again. It does seem to take some time for the misfire to happen. It’s not right when you start the car.
